Ayd применил амитриптилин в больнице Франклин-Сквер в Балтиморе (США) и установил его антидепрессивную активность. Затем данная сфера начала бурно развиваться и появилось значительное количество новых антидепрессантов, но они не компенсировали недостатки первых представителей группы – медленное развитие терапевтического действия, отсутствие эффекта у значительной части пациентов. Накапливались также данные о большом количестве побочных эффектов и противопоказаний. Разработка быстродействующих антидепрессантов для лечения рефрактерных к терапии тяжелых депрессивных или биполярных аффективных расстройств представляет актуальную медицинскую проблему.
