A new analytical method combining solid-phase extraction with high-performance liquid chromatography-tandem mass spectrometry (HPLC-MS/MS) can detect and quantify five amatoxins and two tryptamines (psilocybin and bufotenine) in mushrooms. The method achieved limits of detection as low as 2.0 μg/kg for bufotenine, 5.0 μg/kg for psilocybin, and 10 μg/kg for amatoxins, with good linearity (r² > 0.99) and recoveries ranging from 71.8% to 117%. Applied to 59 wild mushroom samples from Fujian Province, it identified two toxic specimens: Amanita fuligineoides contained α-amanitin (607 mg/kg), β-amanitin (377 mg/kg), and phalloidin (69.0 mg/kg), while a Tricholomataceae sample had psilocybin at 12.6 mg/kg.
