Chromosomal Damage in Human Leukocytes Induced by Lysergic Acid Diethylamide

Lysergic acid diethylamide added to cultured human white blood cells caused a large increase in chromosomal abnormalities. The breaks were not random, with many occurring in chromosome 1. A patient treated with the drug for paranoid schizophrenia over four years also showed similar chromosomal damage.

Lysergic acid diethylamide (LSD-25) can cause chromosomal abnormalities in human leukocytes, as shown by in vitro and in vivo studies. This report extends earlier observations with additional in vitro findings and a larger sample of patients. The study examined peripheral leukocyte cultures from six healthy individuals to assess chromosomal aberrations induced by LSD.

After nearly 40 years of study, the genetic toxicology of lysergic acid diethylamide (LSD) remains inconclusive. In vitro studies suggest LSD either suppresses mitosis or enhances chromosome damage, but these findings do not translate clearly to living organisms. Controlled studies of pure LSD in humans indicate a possible transient increase in lymphocyte chromosome breakage, while animal cytogenetic results are contradictory. LSD is at best a weak mutagen, if mutagenic at all. It is not teratogenic in humans. Three reported leukemia cases among LSD users are likely coincidental, and no clear conclusions about clastogenicity, mutagenicity, teratogenicity, or oncogenicity can be drawn from the available data.
