In patients with treatment-resistant depression receiving esketamine, mystical experiences—similar to those induced by classic psychedelics—occurred in 58% of patients, with high variability across sessions. Higher mean and peak scores on the Mystical Experience Questionnaire (MEQ-30) were associated with greater improvement in depression severity, while dissociative or other non-mystical effects were not. Positive mood and mystical dimensions of the MEQ predicted therapeutic outcomes, and baseline spirituality predicted both treatment response and peak MEQ scores in the first week. These findings suggest that psychedelic-like mystical experiences may contribute to esketamine's therapeutic efficacy.

Contemplative practices like mindfulness and meditation can reduce egoic boundaries, which in turn diminishes existential fears and improves impulse regulation. This conceptual model proposes that enhanced self-control and lower death anxiety are key mechanisms through which these practices foster emotional stability, resilience, and adaptive behavior. The framework integrates contemporary psychological theory with contemplative insights to explain personal growth and spiritual development, offering a basis for future empirical testing and applications in therapy and organizations.

Lysergic acid diethylamide (LSD) loosens the brain's usual alignment between anatomical structure and neural activity in a frequency-dependent way. Low-frequency brain waves (theta, alpha, beta) become less constrained by the structural connectome, indicating a global relaxation of large-scale dynamics. High-frequency gamma activity shows selective reorganization rather than uniform disruption. Greater gamma-band decoupling within core default-mode network regions predicts the intensity of ego dissolution across individuals. LSD does not cause indiscriminate disintegration but drives system-specific rebalancing: visual and attentional systems decouple while auditory networks strengthen coupling. These findings suggest psychedelic states emerge from frequency-dependent relaxation of structural constraints, with default-mode reorganization as a neural correlate of ego dissolution.

Mystical-type experiences induced by psychedelic therapy are commonly linked to reductions in anxiety and depression symptoms, according to a scoping review of 13 clinical trials involving 410 participants. Among studies with life-threatening disease populations, 80% reported a positive relationship between mystical-type experiences and mood improvement, compared to 63% of studies with depressive populations. The review suggests this relationship may depend on factors like timing of symptom assessments and therapeutic context. Future research should examine variables affecting mystical-type experiences and other aspects of set and setting to optimize positive outcomes.
